Avabopa(avatrombopag) is indicated for the treatment of thrombocytopenia in adult patients in the following two conditions: 1.Patients with Chronic Liver Disease (CLD): Used to increase platelet counts prior to a scheduled medical procedure that carries a risk of bleeding. 2.Patients with Chronic Immune Thrombocytopenia (ITP): Used in patients who have had an insufficient response to prior therapy (e.g., corticosteroids, immunoglobulins) to increase and maintain platelet counts.

Indications

Avabopa(avatrombopag) is indicated for the treatment of thrombocytopenia in adult patients in the following two conditions:

  1. Patients with Chronic Liver Disease (CLD): Used to increase platelet counts prior to a scheduled medical procedure that carries a risk of bleeding.

  2. Patients with Chronic Immune Thrombocytopenia (ITP): Used in patients who have had an insufficient response to prior therapy (e.g., corticosteroids, immunoglobulins) to increase and maintain platelet counts.



Mechanism of Action

Avabopa(avatrombopag) is a thrombopoietin (TPO) receptor agonist. Its active substance, avatrombopag, binds to the human TPO receptor and mimics the biological activity of endogenous TPO, thereby stimulating the proliferation and differentiation of megakaryocytes in the bone marrow, leading to increased platelet production.


Dosage and Administration

  • For Patients with Chronic Liver Disease (pre-procedure):

    • Dose: 40 mg (2 tablets) or 60 mg (3 tablets) once daily for 5 consecutive days.

    • Timing: Dosing should start 10 to 13 days prior to the planned procedure. The procedure should be scheduled to occur 5 to 8 days after the last dose.

    • Administration: Take once daily with food.

  • For Patients with Chronic Immune Thrombocytopenia (ITP):

    • Starting Dose: The usual starting dose is 20 mg (1 tablet) once daily.

    • Dose Adjustment: The dose will be adjusted based on regular monitoring of platelet counts by a physician.

    • Administration: Take once daily with food.



Contraindications and Important Precautions

  • Contraindications: Hypersensitivity to avatrombopag or any of the excipients.

  • Thromboembolic Risk: This medicine may increase the risk of arterial and venous thrombotic events. Use with caution in patients with known risk factors for thrombosis (e.g., older age, prolonged immobilization, cancer, smoking, obesity) and monitor for signs and symptoms of thrombosis.

  • Bone Marrow Examination: Treatment may worsen pre-existing bone marrow conditions such as Myelodysplastic Syndromes (MDS). Monitoring and bone marrow examination may be necessary.

  • Special Populations: Not recommended during pregnancy. Breast-feeding women should consult their doctor to weigh the benefits against potential risks. Not recommended for use in children and adolescents under 18 years of age.

  • Lactose: This medicine contains lactose. Patients with lactose intolerance or other sugar metabolism issues should use with caution.



Storage

Store at room temperature. Keep out of the sight and reach of children.
